Fluorescent Hyaluronic Acid Methacryloyl(Green fluorescent HAMA)

Hyaluronic acid methacryloyl (HAMA) is a double-bonded modified hyaluronic acid, which can be crosslinked to form hydrogels by UV and visible light under the addition of photoinitiators.HAMA hydrogels have excellent biocompatibility and adjustable mechanical properties. Fluorescent HAMA is chemically grafted with fluorescent molecules on HAMA molecules, which can be made to have specific fluorescent colors by changing the type of fluorescent molecules. This chemical labeling method avoids the drawbacks of physical mixing or electrostatic adsorption where the fluorescent molecules tend to diffuse out of the system, and also avoids the drawbacks of uneven imaging of fluorescent particles, which is widely used in biomedical fields such as tissue engineering, 3D printing, and drug delivery.

Steps for use:

1. Preparation of 0.5mg/mL initiator standard solution

Add 25mL of solvent (sterilized water, PBS, etc.) to the brown glass vial containing the initiator LAP and dissolve for 30min at room temperature, shaking several times to fully dissolve.

2. Prepare fluorescent HAMA solution (recommended concentration 2-10%)

Take appropriate amount of fluorescent HAMA into the centrifuge tube, add the prepared LAP standard solution, dissolve for 30min at room temperature, oscillate several times and fully dissolve.


Curing into gel:

1. Use 405 nm blue light to irradiate the above solution containing the initiator, about 30s can be cured into glue (irradiation time is related to the concentration of fluorescent HAMA and blue light source, according to the required strength of the glue can be adjusted to the appropriate curing time);

2. if use 2959 initiator, need to use 365nm UV irradiation and extend the irradiation time (LAP is more efficient than 2959 initiator, shorter time), UV will hurt the skin and eyes, pay attention to protect.


Precautions:

1. It is recommended to increase the irradiation time to get better curing effect. Recommended time: fluorescent HAMA concentration ≤5%, irradiation time ~ 30 s; fluorescent HAMA concentration >5%, irradiation time ~ 20 s. 2;

2. Use 2959 initiator needs higher temperature to dissolve in water or PBS (solubility in water ≥ 5 mg/mL);

3. LAP initiator solution can be stored for 12 months at 4°C protected from light, but it is recommended to use it now;

4. It is recommended to use LAP initiator for cellular experiments because blue light irradiation is gentler to cells than UV.

5. After adding LAP, the solution needs to be protected from light and the centrifuge tube can be wrapped with tin foil;
