Molecular Structure:
Steps for use:
1. Preparation of 0.5 mg/mL initiator standard solution
Add 20 mLPBS to the brown glass bottle containing the initiator LAP, and dissolve for 30 min in a 37 ℃ water bath with repeated shaking to fully dissolve.
2. Prepare AlgMA solution (recommended concentration 4-10%)
Take appropriate amount of AlgMA into a centrifuge tube, add the prepared LAP standard solution, dissolve at room temperature for 0.5-1h, shaking several times, fully dissolved.
Curing into gel:
1. Use 405nm blue light to irradiate the above solution containing initiator, about 30s can be cured into glue (irradiation time is related to the concentration of AlgMA and the blue light source, according to the required strength of the glue can be adjusted to the curing time);
2. if use 2959 initiator, need to use 365nm UV irradiation and extend the irradiation time (LAP is more efficient than 2959 initiator, shorter time), UV light will hurt the skin and eyes attention to protection.
Precautions:
1. It is recommended to increase the irradiation time to get better curing effect. Recommended time: AlgMA concentration ≤ 5%, irradiation time ~ 40s; AlgMA concentration > 5%, irradiation time ~ 30s. 2;
2. The use of 2959 initiator requires higher temperature to dissolve in water or PBS (solubility in water ≥ 5 mg/mL);
3. LAP initiator solution can be stored for 12 months at 4℃ protected from light, but it is recommended to use it now;
4. It is recommended to use LAP initiator for cellular experiments, blue light irradiation is gentler to cells than UV;
5. If ionic cross-linking of AlgMA is required, it is recommended to use deionized water instead of PBS to prepare the standard solution of initiator. ( PO43- ions in PBS will produce Ca3 ( PO4 ) 2 precipitation with Ca2+ ions, which will make the gel opaque, in addition, the monovalent ions in PBS will compete with the Ca2+ ions of AlgMA, which will make the ionic cross-linking weaker or even no ionic cross-linking;)
6. After adding LAP, the solution should be protected from light, and the centrifuge tube can be wrapped with tin foil.
